Title Light pollution may create demographic traps for nocturnal insects Type Journal Article

Abstract Light pollution impacts both intra- and inter-specific interactions, such as interactions between mates and predator–prey interactions. In mobile organisms attracted to artificial lights, the effect of light pollution on these interactions may be intensified. If organisms are repelled by artificial lights, effects of light pollution on intra- and inter-specific interactions may be diminished as organisms move away. However, organisms repelled by artificial lights would likely lose suitable habitat as light pollution expands. Thus, we investigated how light pollution affects both net attraction or repulsion of organisms and effects on intra- and inter-specific interactions. In manipulative field studies using fireflies, we found that Photuris versicolor and Photinus pyralis fireflies were lured to artificial (LED) light at night and that both species were less likely to engage in courtship dialogues (bioluminescent flashing) in light-polluted field plots. Light pollution also lowered the mating success of P. pyralis. P. versicolor is known to prey upon P. pyralis by mimicking the flash patterns of P. pyralis, but we did not find an effect of light pollution on Photuris–Photinus predator–prey interactions. Our study suggests, that for some nocturnal insects, light-polluted areas may act as demographic traps, i.e., areas where immigration exceeds emigration and inhibition of courtship dialogues and mating reduces reproduction. Examining multiple factors affecting population growth in concert is needed to understand and mitigate impacts of light pollution on wildlife.

Abstract The awareness of the interrelationship between immunosenescence and constant light exposure can provide new insights into the consequences of excessive exposure to light at night due to light pollution or shift work. Here, we investigated whether constant light exposure (LL) acts as an inducer of immunosenescence. We also determined the role of melatonin or turmeric in reversing the putative effects of constant light and explored for the first time the underlying molecular mechanisms. Young (3-4-month-old) rats were exposed daily to LL alone or in combination with each of melatonin and turmeric for 12weeks. A group of aged rats (18-months old; n=6) was used as a reference for natural immunosenescence. Constant light exposure resulted in remarkable pathophysiological alterations resembling those noticed in normal aged rats, manifested as apparent decreases in antioxidant activities as well as Nrf2 and DJ-1 expressions, striking augmentation in oxidative stress, proinflammatory cytokines and expression of TNFalpha, Bax, and p53 genes, and deleterious changes of lymphoid organs, Co-administration of melatonin or turmeric was able to reverse all alterations induced by LL through upregulation of Nrf2/DJ-1 and downregulation of p53/Bax pathways. These data suggest that LL accelerates immunosenescence via oxidative stress and apoptotic pathways. They also demonstrate for the first time that turmeric is comparable to melatonin in boosting the immune function and counteracting the LL-associated immunosenescence. These effects suggest that turmeric supplementation can be used as an inexpensive intervention to prevent circadian disruption-related immunosenescence. However, to validate the effects of turmeric on humans further studies are warranted.

Title Impact of a shift work-like lighting schedule on the functioning of the circadian system in the short-lived fish Nothobranchius furzeri Type Journal Article

Abstract Adult Nothobranchius furzeri of the MZM-04/10 strain were individually kept and subjected to a “5+2” shifting lighting schedule (SHIFT) for 8weeks in order to evaluate the desynchronizing effects of a simulated human-like shift-work schedule on the functioning of the circadian system (CS). With this aim, sixteen 21-week-old N. furzeri were placed into a Morning, Night and Evening schedule (lights on from 08:00 to 16:00, 00:00 to 08:00 and 16:00 to 00:00h, respectively) and fed once a day in the middle of the corresponding photophase (12:00, 04:00 and 20:00h, respectively). Then, in the weekends (2days), fish were always returned to the Morning shift. As controls, 16 fish were maintained under a non-shifting LD cycle condition (CONTROL) throughout the whole experiment, with lights on from 08:00 to 16:00h. Rest-activity rhythm (RAR) of fish subjected to SHIFT showed several symptoms of chronodisruption, such as a decrease in the percentage of diurnal activity and a reduction of the relative amplitude and the circadian function index with time. When a periodogram analysis was performed, RAR of N. furzeri under SHIFT conditions showed up to three separate circadian components: one longer than 24h (26.5h) that followed the weekly 8h delays; a short-period component (~23h) that was related to the weekend's phase advances, and finally, a 24h component. The shifting LD schedule also affected fish CS at a molecular level, with several significant differences in the expression of core genes of the molecular clock (bmal1, clock, roralpha, rev-erbalpha) between SHIFT and CONTROL animals. RAR impairment along with changes in clock gene expression could be associated with high stress and accelerated aging in these fish.

Title Regulation of the MAPK signaling pathway by miR-421-5p in rats under light pollution Type Journal Article

Abstract The present study aimed to explore the difference in the expression profiles of ovarian microRNA sequences in rats in a light pollution environment and rats in a normal light environment. Rats in the control group were exposed to 12h light/dark cycles, while rats in the model group were continuously exposed to 24h light. The ovaries were extracted from the two groups of rats, and Illumina HiSeq 2500 highthroughput sequencing technology was used to detect the differences in microRNA (miRNA) expression among the two groups. Fluorescence quantitative reverse transcriptionpolymerase chain reaction was used to verify the differential expression of miRNA. The present study was designed to experimentally validate the interaction between miR4215p and mitogenactivated protein kinase (MAPK) 7 by using the dualluciferase reporter system, and to explore the expression of proteins in the MAPK signaling pathway with a lentiviral vectormediated small hairpin RNA interference against microRNA4215p. The expression of 45 miRNAs was significantly different. In total, 13 miRNAs were upregulated, of which 5 miRNA sequences were known and 8 were predicted. Furthermore, 32 miRNAs were downregulated, of which 11 miRNA sequences were known and 21 were predicted. The results of the luciferase reporter assay confirmed the targeting association between miR4215p and MAPK7. The expression levels of MAPK and genes in its downstream signaling pathways, including cFos, CREB and cMyc, were downregulated when miR4215p was overexpressed and upregulated when miR4215p was silenced. The differential expression of miRNAs may serve an important role in the development of the ovary in a light pollution environment. miR4215p may regulate ovarian growth and development by targeting the MAPK signaling pathway in light polluted rat ovaries.

Abstract Light pollution or artificial light at night (ALAN) is an increasing, worldwide challenge that affects many aspects of animal behaviour. Interestingly, the response to ALAN varies widely among individuals within a population and variation in personality (consistent individual differences in behaviour) may be an important factor explaining this variation. Consistent individual differences in exploration behaviour in particular may relate to the response to ALAN, as increasing evidence indicates its relation with how individuals respond to novelty and how they cope with anthropogenic modifications of the environment. Here, we assayed exploration behaviour in a novel environment as a proxy for personality variation in great tits (Parus major). We observed individual sleep behaviour over two consecutive nights, with birds sleeping under natural dark conditions the first night and confronted with ALAN inside the nest box on the second night, representing a modified and novel roosting environment. We examined whether roosting decisions when confronted with a camera (novel object), and subsequently with ALAN, were personality-dependent, as this could potentially create sampling bias. Finally, we assessed whether experimentally challenging individuals with ALAN induced personality-dependent changes in sleep behaviour.

Slow and fast explorers were equally likely to roost in a nest box when confronted with either a camera or artificial light inside, indicating the absence of personality-dependent sampling bias or avoidance of exposure to ALAN. Moreover, slow and fast explorers were equally disrupted in their sleep behaviour when challenged with ALAN. Whether other behavioural and physiological effects of ALAN are personality-dependent remains to be determined. Moreover, the sensitivity to disturbance of different behavioural types might depend on the behavioural context and the specific type of challenge in question. In our increasingly urbanized world, determining whether the effects of anthropogenic stressors depend on personality type will be of paramount importance as it may affect population dynamics.
